Spectral-domain differential interference contrast microscopy.

چکیده
We present a fiber-optic low-coherence imaging technique, termed spectral-domain differential interference contrast microscopy (SD-DIC), for quantitative DIC imaging of both reflective surfaces and transparent biological specimens. SD-DIC combines the common-path nature of a Nomarski DIC interferometer with the high sensitivity of spectral-domain low-coherence interferometry to obtain high-resolution, quantitative measurements of optical pathlength gradients from a single point on the sample. Full-field imaging can be achieved by scanning the sample. A reflected-light SD-DIC system was demonstrated using a USAF resolution target as the phase object. Live cardiomyocytes were also imaged, achieving a resolution of 36 pm for pathlength gradient measurements. The dynamics of cardiomyocyte contraction were recorded with high sensitivity at selected sites on the cells.

High-resolution differential interference contrast X-ray zone plates: Design and fabrication
Differential interference contrast is a potentially powerful technique for contrast enhancement in soft X-ray microscopy. We describe the design and fabrication of single-element diffractive optical elements suitable as objectives for high-resolution differential interference contrast microscopy in the water-window spectral range. A one-dimensional pattern calculation followed by an extension t...
متن کاملReconstruction of optical pathlength distributions from images obtained by a wide-field differential interference contrast microscope.
An image processing algorithm is presented to reconstruct optical pathlength distributions from images of nonabsorbing weak phase objects, obtained by a differential interference contrast (DIC) microscope, equipped with a charge-coupled device camera. The method is demonstrated on DIC images of transparent latex spheres and unstained bovine spermatozoa. The images were obtained with a wide-fiel...
متن کاملMultimodal spectral imaging of cells using a transmission diffraction grating on a light microscope.
A multimodal methodology for spectral imaging of cells is presented. The spectral imaging setup uses a transmission diffraction grating on a light microscope to concurrently record spectral images of cells and cellular organelles by fluorescence, darkfield, brightfield, and differential interference contrast (DIC) spectral microscopy. Initially, the setup was applied for fluorescence spectral i...
متن کاملMotion-enhanced, differential interference contrast (MEDIC) microscopy of moving vesicles in live cells: VE-DIC updated.
Video-enhanced differential interference contrast microscopy with background subtraction has made visible many structures and processes in living cells. In video-enhanced differential interference contrast, the background image is stored manually by defocusing the microscope before images are acquired. We have updated and improved video-enhanced differential interference contrast by adding auto...

Progress in lithography and nanofabrication @E. Di Fabrizio et al., Nature ~London! 401, 895 ~1999!# has made it possible to apply differential interference contrast ~DIC! in x-ray microscopy using an original x-ray doublet lens based on two specially developed zone plates. Switching from bright-field imaging ~absorption contrast! to x-ray DIC, we observe, similar to visible-light microscopy, a...
